dc.description.abstractThis document is the examination paper for the MIC 311 course, "Microbial Physiology and Metabolism," at the University of Fort Hare. The main examination took place in May/June 2025 and is worth 100 marks, with a duration of 3 hours. The paper consists of four questions and requires students to answer all of them. The first question covers the history of cell discovery and the differences between the plasma membrane and the outer membrane of Gram-negative bacteria. The second question focuses on microbial growth, requiring a comparison of growth phases in batch culture and a discussion on how to maintain a bacterial culture in the exponential phase for an industrial process. The third question addresses the classification of prokaryotes based on energy and carbon requirements and requires a derivation of the bacterial generation time equation. The final question asks for a discussion of the differences between Gram-positive and Gram-negative bacteria cell walls, a description of three methods for measuring cell numbers (including their applications, disadvantages, and advantages), and the identification of three factors that limit microbial growth in a batch culture.
